A Comprehensive Guide to the Light Microscope

1. Operation Guide

(1) Brightfield Observation
Plug in the microscope's power cord. Turn on the brightfield power switch. Then, adjust the brightness knob to a suitable level.

●Adjust the position of the phase contrast plate, and place it at the center light-passing hole.
●Place the specimen slide or petri dish. Select the required petri dish holder based on the outer size of the petri dish. Adjust the X/Y axis handwheels to move the specimen or petri dish to the center of the field of view.
●Rotate the nosepiece to select an objective lens, and adjust the aperture diaphragm to achieve suitable field brightness.
●Adjust the coarse and fine focusing knobs to get a clear image.
●After observation, turn off the microscope power switch. If you do not use it for a long time, I recommend unplugging the power cord.

(2) Phase Contrast Observation
●Under brightfield conditions, place the sample and focus it, following the same steps as the brightfield observation above.
●Switch to phase contrast objectives (objectives marked with "ph").
●Based on the magnification of the phase contrast objective, adjust the phase contrast ring plate. The 10X phase ring works with the 10X phase objective. The 20X phase ring matches the 20X and 40X objectives.
When using the 4X objective, place the phase contrast ring plate in the center. Also, reduce the size of the condenser diaphragm.
●Adjust the coarse and fine focusing knobs to get a clear image.

(3) Fluorescence Observation
●Turn off the brightfield light source, and plug in the independent power cord of the LED fluorescence accessory.
●Place the sample at the center of the optical path.
●Rotate the objective turret to select the objective lens for observation.
Turn on the fluorescence module switch. Select the desired fluorescence channel (B/G/U) using the lever or knob. Adjust the brightness to the right level.

●Use the coarse and fine focusing knobs to get a clear image. When you switch fluorescence channels or objectives, the sample image might be a bit out of focus. Use the fine focus to make it clearer.
